The company said electricity demand across Scotland is expected to double by 2050 and the "essential" upgrades will create additional capacity.
This will enable more renewable energy to be transported efficiently from where it is generated to homes and businesses across the country.
Ian Page, project manager for the Mossmorran substation upgrade, said: “Following the previous consultation, we are giving the community a further opportunity to review the revised proposals and make comments before we finalise the design that we will submit for planning consent.
“Local feedback is a crucial part of developing our designs and it is important that we listen and take this on board where we can, to create the optimal design for the expansion of Mossmorran facility and get more clean, green electricity into the grid network.”
There will be two consultations, giving local residents the chance to meet the project team, view the updated plans and ask questions.
The first drop-in event is on Tuesday June 23 from 2pm to 7pm in Auchtertool Village Hall.
The second is the following day, Wednesday June 24, between 11am and 4pm in Crossgates Royal British Legion, which is on Main Street.
Cowdenbeath MSP David Barratt said: "I know this remains of significant interest for many local residents, particularly those who may be directly affected by the proposals.
"It is important that people have a chance to review the plans and make their views known."
The substation is near to the proposed site for a massive 600 megawatts data centre, which has already prompted significant opposition and could be built on land north-west of Auchtertool.
ILI Cato Limited is seeking planning permission in principle for the development, which could have a complex of data buildings up to 35 metres in height.
SP Energy Networks said their proposals are part of a long‑term investment to unlock regional economic growth, increase capacity and ensure a secure and reliable power supply for future generations.
The company said feedback from local communities will help shape the final design for the substation layout, access, drainage and landscaping.

The company stated: "Much of Scotland’s transmission network was built between 50 and 100 years ago.
"While it has evolved over time, sections of the network are now nearing operational limits at a time when more wind, solar and battery storage projects are seeking to connect.
"To accommodate this growth, the voltage of overhead lines between Tealing and Kincardine will increase from 275kV to 400kV.
"This requires upgrades at Mossmorran and Westfield substations, along with a new substation at Conland, north of Leslie."
